Ehbp1

E8.5 Ehbp1^-/- embryos are sometimes smaller than litter mates but all fall within a normal range for their litters. By E9.5 almost all mutants are incompletely turned (n=10/12) and have abnormally shaped heads (particularly forebrain and midbrain; n=10/12). A majority display kinks in the caudal neural tube (n= 8/12), abnormally shaped hearts (n= 8/12), branchial arch defects (n=8/12) or abnormal tailbud (n= 7/12). A minority of mutants also display incomplete cranial neural tube closure (n= 5/12) and some have pale yolk sacs (n=3/12). At E10.5 (not depicted) all Ehbp1 KO conceptus’ have pale yolk sacs and many are dying.

At E9.5, all Ehbp1^-/- placentas examined (n=3) are properly developed and comparable to controls. A slight reduction in the Tfeb+ and Gcm1+ trophoblasts within the labyrinth zone in all Ehbp1 KO placentas, is likely due to mild developmental delay of the conceptus compared to control littermates.

E8.5 Ehbp1 mutants sectioned and examined by IF (n=3) display appropriate expression and patterning of FoxA2, SMA and Sox2, demonstrating that the germ layers have formed.

By E9.5 all Ehpb1^-/- embryos examined (n=3) display appropriate germ layer patterning. The entire mutant embryo is contorted by the unturned tail, which is observed in the H & E stained section on the right.
