Traf7 -/- embryos display a normal phenotype at E8.5. At E9.5 Traf7 -/- embryos are mainly normal although a fraction are delayed, some display incomplete turning and others have abnormal blood deposition in the embryo or in the yolk sac. At E10.5 almost all Traf7 -/- embryos are delayed with abnormal head shape, and most have abnormal cardiac tissue, some with enlarged hearts and some with pericardial edema. Most mutants display abnormal branchial arch development. Mutants often display abnormal blood deposition. The mutant yolk sacs that were dissected intact at E10.5 are vascularized but appear paler than their control littermates.

At E9.5, Traf7-/- placentas are properly developed and comparable to controls.

At E10.5, Traf7-/- placentas exhibit multiple labyrinth zone defects compared to control littermates. In KO placentas, the Tfeb+ labyrinth trophoblast layer is reduced in thickness but displays elevated Tfeb signal intensity, suggesting an increased density of Tfeb+ cells. Chorioallantoic branching is nearly absent, as Gcm1 expression is sparse and remains at the chorioallantoic interface.

Traf7 -/-embryos display appropriate germ layer formation at E9.5-10.5. At E9.5 Traf7 -/-embryos are mainly normal, displaying normal heart and liver development. By E10.5 Traf7 -/- embryos are delayed and section analysis demonstrates a reduction in cellular density which is most easily observed when comparing the heads sections to controls which are less cellularly dense. E10.5 Traf7 -/- embryos also display abnormal blood deposition, as observed in the midbrain and the pericardial sac of the mutant displayed above.

In situ hybridization performed on whole WT embryos from E8.5-10.5 demonstrates relatively ubiquitous expression of Traf7 (purple/blue color) at all stages examined.

Section in situ hybridization is used to detect Traf7 expression (purple/blue) in WT E8.5-10.5 placentas. Traf7 is widely expressed in the fetal compartment of the placenta (junctional and labyrinth zone) at E8.5. By E9.5, Traf7 expression becomes enriched in the spongiotrophoblast layer and in specific cells within the labyrinth zone. This pattern of expression remains at E10.5. These sections have been counterstained with eosin.
